چکیده
1. Methylxanthines (IBMX, caffeine, theophylline), which are known inhibitors of c-AMP phosphodiesterase inhibited reversibly progesterone-induced maturation of Xenopus oocyte. 2. The time course of protein phosphorylation during maturation stimulated by progesterone was analysed in Xenopus oocyte : protein phosphorylation increased around germinal vesicle breakdown (GVBD). No quantitative changes were observed before this period. 3. Injection of the heat-stable inhibitor (I) of c-AMP-dependent protein kinases into Xenopus and Discoglossus oocytes triggered GVBD with 30-50 p. 100 efficiency. When (I)

O-10: A Marked Animal-Vegetal Polarity in The Localization of Na+,K+-ATPase Activity and Its Down-Regulation Following Progesterone-Induced Maturation
Background: Polarized cells are key to the process of differentiation. Xenopus oocyte is a polarized cell that has complete blue-print to differentiate 3 germ layers following fertilization, as key determinant molecules (Proteins and RNAs) are asymmetrically localized. The objective of this work was to localize Na+, K+-ATPase activity along animal-vegetal axis of polarized Xenopus oocyte and fo...
متن کاملP-217: The Effect of Progesterone on The In Vitro Maturation of Mouse Germinal Vesicle Oocytes Derived from Polycystic Ovary
Background: Polycystic ovary syndrome (PCOS) is characterized by chronic anovulation and hyperandro genism and affects approximately 5-10% of women of reproductive age. In vitro maturation (IVM) of mammalian oocytes has become an efficient method to produce mature oocytes in order to use in assisted reproductive techniques such as in vitro fertilization (IVF), intra cytoplasmic sperm injection ...
متن کاملInhibition of mos-induced oocyte maturation by protein kinase A
The relationship between the mos protooncogene protein and cAMP-dependent protein kinase (PKA) during the maturation of Xenopus oocytes was investigated. Microinjection of the PKA catalytic subunit (PKAc) into Xenopus oocytes inhibited oocyte maturation induced by the mos product but did not markedly affect the autophosphorylation activity of injected mos protein. By contrast, PKAc did not inhi...
متن کاملProgesterone inhibits protein kinase A (PKA) in Xenopus oocytes: demonstration of endogenous PKA activities using an expressed substrate.
3'-5' cyclic adenosine monophosphate (cAMP)-dependent protein kinase, PKA, is thought to be a key enzyme that controls prophase arrest in vertebrate oocytes. It has long been established that overexpression of the catalytic subunit of PKA inhibits hormone-induced frog oocyte maturation whereas overexpression of the regulatory subunits induces hormone-independent oocyte maturation. However, the ...
متن کاملPossible involvement of phosphatidylinositol 3-kinase, but not protein kinase B or glycogen synthase kinase 3beta, in progesterone-induced oocyte maturation in the Japanese brown frog, Rana japonica.
It is known that amphibian oocytes undergo maturation through the formation and activation of maturation-promoting factor (MPF) in response to stimulation by the maturation-inducing hormone progesterone; however, the signal transduction pathway that links the hormonal stimulation on the oocyte surface to the activation of MPF in the oocyte cytoplasm remains a mystery. The aim of this study was ...
